RESUMO

The purpose of this study was to investigate whether a defensin-like antimicrobial peptide (C-13326 peptide) identified in Hermetia illucens could possess protective effect against multidrug-resistant Aeromonas schubertii in hybrid snakehead (Channa maculate ♀ × Channa argus ♂). The cDNA of C-13326 peptide comprised 243 nucleotides encoding 80 amino acids, with six conserved cysteine residues and the classical CSαß structure. The recombinant expression plasmid pPIC9K-C-13326 was constructed and transformed into GS115 Pichia pastoris, and the C-13326 peptide was expressed by induction with 1% methanol. The crude extract of C-13326 peptide was precipitated by ammonium sulfate, assayed by Braford method, detected by tricine-SDS-PAGE, evaluated by BandScan software and identified by liquid chromatography-mass spectrometry. The C-13326 peptide was shown to have inhibitory activity against the growth of multidrug-resistant A. schubertii DM210910 by using the minimum growth inhibitory concentration and Oxford cup method. In addition, scanning electron microscopy analysis suggested that C-13326 peptide inhibited the growth of A. schubertii DM210910 by damaging the bacterial cell membrane. To explore the role of peptide C-13326 in vivo, hybrid snakehead was fed with peptide C-13326 as feed additives for 7 days. The results revealed that C-13326 peptide could significantly down-regulate the expression levels of IL-1ß, IL-8, IL-12 and TNF-α (p < .05), and significantly improved the survival rate of hybrid snakehead after challenging with A. schubertii DM210910. Therefore, the C-13326 peptide is a promising antimicrobial agent for A. schubertii treatment in aquaculture.

RESUMO

Multipartite entanglement is one of the most fundamental and important resources for quantum information processing in both discrete variable and continuous variable (CV) regimes. For its applications in the CV regime, such as the realization of quantum teleportation networks and quantum dense coding, the quadrature squeezing of multipartite entanglement plays a significant role. Here, we report the first, to the best of our knowledge, experimental measurement of the quadrature squeezing in the quadripartite entanglement generated by the two-beam pumped cascaded four-wave mixing process in a 85 R b vapor cell. Moreover, we find that the quadrature squeezing is nonexistent in each pair of beams, but exists in the whole quadripartite entanglement. Our results may find potential applications in building a multi-user quantum secret sharing network.

RESUMO

Named for the characteristic basic helix-loop-helix (bHLH) region in their protein structure, bHLH proteins are a widespread transcription factor class in eukaryotes. bHLHs transcriptionally regulate their target genes by binding to specific positions on their promoters and thereby direct a variety of plant developmental and metabolic processes, such as photomorphogenesis, flowering induction, shade avoidance, and secondary metabolite biosynthesis, which are important for promoting plant tolerance or adaptation to adverse environments. In this review, we discuss the vital roles of bHLHs in plant responses to abiotic stresses, such as drought, salinity, cold, and iron deficiency. We suggest directions for future studies into the roles of bHLH genes in plant and discuss their potential applications in crop breeding.

RESUMO

We give the general expressions of intensity-difference squeezing (IDS) generated from two types of optical parametric amplifiers [i.e. phase-sensitive amplifier (PSA) and phase-insensitive amplifier (PIA)] based on the four-wave mixing process, which clearly shows the IDS transition between the ultra-low average input photon number regime and the ultra-high average input photon number regime. We find that both the IDS of the PSA and the IDS of the PIA get enhanced with the decrease of the average input photon number especially in the ultra-low average input photon number regime. This result is substantially different from the result in the ultra-high average input photon number regime where the IDS does not vary with the average input photon number. Moreover, under the same intensity gain, we find that the optimal IDS of the PSA is better than the IDS of the PIA in the ultra-low average input photon number regime. Our theoretical work predicts the presence of strong quantum correlation in the ultra-low average input photon number regime, which may have potential applications for probing photon-sensitive biological samples.

RESUMO

OBJECTIVE: To investigate the intervention effects and mechanisms of the compound panax notoginsenoside granules (CPNG) on bleomycin-induced pulmonary fibrosis in rats. METHODS: Sixty male Sprague-Dawley rats were randomly divided into 6 groups, including normal control group, model group, prednisolone group, and CPNG (high, middle and low-dose) groups. Each group included 10 Sprague-Dawley rats. Except the control group, pulmonary fibrosis model rats were injected with bleomycin (5 mg/kg) via trachea. On the second day of the bleomycin injection, the rats in each treatment group were given prednisolone (3.33 mg/kg) and corresponding doses of CPNG (100, 50, 25 mg/kg) by nasogastric feeding daily; the normal control group and the model group were given the equal volume distilled water daily. Twenty-eight days later, all rats were executed and the lung tissues were subjected to the histopathological examination via HE and Masson staining. The hydroxyproline (HYP) in lung tissues was assessed by alkaline hydrolysis. The expressions of TGF-ß1, Smad2, and Smad7 proteins were analyzed by Western blotting. RESULTS: Compared with the model group, the histopathological results revealed that CPNG dramatically decreased the severity of lung inflammation and fibrosis in rats (P<0.01). Compared with the control group, the HYP in lung tissues was reduced in all of the CPNG groups (P<0.01). Moreover, CPNG inhibited the expressions of TGF-ß1 or Smad2 proteins (P<0.01), but enhanced the expression of Smad7 protein in the rat lung tissues (P<0.01). CONCLUSION: CPNG has the prevention and treatment effect on the bleomycin-induced pulmonary fibrosis in rats, and it might be attributed to the regulation on the TGF-ß1/Smads signaling pathway.
